چکیده

Diffusion of aqueous sodium dodecyl sulfate (SDS) across cross-linked polyacrylamide hydrogel membranes has been studied by electrical conductivity measurements. Initial rapid sorption of SDS (as unimer) into the membranes is observed. The effect of SDS concentration, and of cross-linker fraction on the degree of swelling of the gels is studied and associated with binding of the surfactant to the polymer, with surface bound water suggested to be involved in these interactions. Below the surfactant critical micelle concentration, volume collapse of less cross-linked membranes is observed, and associated with aggregate formation. Fluorescence measurements using pyrene as a probe show that micellar aggregates do not diffuse through the membrane, and only overall unimer diffusion is observed. The effect of cross-linking on the diffusion process is discussed. 2002 Elsevier Science Ltd. All rights reserved.

Characterization of fragment E from fibrinogen and cross-linked fibrin.

Fragment E, a terminal plasmin digestion product of fibrinogen or fibrin, contains portions of the alpha, beta, and gamma chains linked by disulfide bonds. In this study, Fragment E from fibrinogen and fully cross-linked fibrin were purified by gel filtration of the soluble fraction from heated plasmin digests of either fibrinogen or fibrin or by step-wise chromatography of terminal plasmin dig...

Ferredoxin Cross-Links to a 22 kD Subunit of Photosystem I.

We have used a cross-linking approach to study the interaction of ferredoxin (Fd) with photosystem I (PSI). The cross-linking reagent N-ethyl-3-(3-dimethylaminopropyl) carbodiimide was found to cross-link spinach Fd to a 22 kilodalton subunit of PSI in both isolated spinach (Spinacia oleracea) PSI complexes and spinach thylakoid membranes. The product had an apparent molecular weight of 38 kilo...

Characterization of Edible Film Fabricated with Channel Catfish Ictalurus punctatus Gelatin by Cross-Linking with Transglutaminase

The objectives of this research were to improve the film-forming properties of Channel Catfish Ictalurus punctatus skin gelatin (CSG) by cross-linking with transglutaminase (TG), determine and optimize the TG reaction time, and characterize the mechanical and barrier properties of CSG edible film. Cross-linking of CSG was performed by TG for 0, 10, 20, 30, and 40 min at 50°C, and the reaction w...

Affinity cross-linking of atrial natriuretic factor to its receptor in bovine adrenal zona glomerulosa.

125I-Labeled atrial natriuretic factor (ANF) was covalently cross-linked to its binding sites in bovine adrenal zona glomerulosa membranes using the hydrophilic cross-linker bis(sulfosuccinimidyl) suberate. Analysis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis in the presence of 2-mercaptoethanol revealed that two protein bands with apparent Mr 68,000 and 114,000 were specifical...

Affinity labeling of the sialoglycopeptide antimitogen receptor.

An 18-kDa 125I-sialoglycopeptide growth inhibitor was covalently cross-linked to its binding site on intact cultured Swiss 3T3 cells by three bifunctional cross-linkers with short (dimethyl adipimate), medium (disuccinimidyl suberate), and long (bis(2-succinimidooxycarbonyloxyethyl)sulfone) chain lengths. Analysis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and autoradiography ...
